Unlike the first TV Series, Fullmetal Alchemist: Brotherhood covers every one of the 27 volumes of the manga. The Elric brothers, Edward and Alphonse were abandoned by their father and were left on their mother's care, Trisha Elric. She then dies from a mysterious disease and the brothers vow to revive her using alchemy. Since their mother's death, Winry and Pinako cared for the boys. Edward and Alphonse learned Alchemy from their father's books and trained for several years with Izumi Curtis just to perform the human transmutation, a ritual considered as taboo by many alchemists.
Their attempt to revive Trisha goes wrong, Alphonse loses his body and Ed loses his left leg. In despair to keep his brother alive, Ed sacrificed his right arm in the process and seals Alphonse's soul in an armor. This events attracted the attention of Roy Mustang. He invited Ed to join the State Alchemists group, which is part of the nation's army. After Ed received his automail prosthesis from Winry, the Elric brothers decided to burn down their home and leave the town for Ed to take the examination to become State Alchemists.
When he passed the exam, Edward earned the title "Fullmetal Alchemist" from the State Military, and the siblings start their journey to find the way of the famous Philosopher's Stone, under the course of Colonel Roy Mustang. They are after the Philosopher's Stone, a stone made of human souls that can give an alchemist great amount of power, so that they can break the the rule of Equivalent Exchange and retrieve back Alphonse's body and Ed's lost body parts. In the process, they discover a plot by a group of alchemy-created human bodies who are plotting to sacrifice the entire population of Amestris to create a Philosopher's stone. They reject the idea of using human souls to achieve their goals. As the story progresses, they learned the conspiracy within the government and reunited with their father who later on reveals his connection with the Homonculi and with the help of Mustang and fellow state alchemist save the lives of people of Amestris.
